چکیده

A differential-type amperometric biosensor based on conventional thick-®lm technology has been developed for breath alcohol measurement. The amperometric breath alcohol biosensor utilizes the alcohol dehydrogenase (ADH) and nicotinamide adenine dinucleotide (NAD‡) cofactor which produce reduced NADH as a product of the oxidation of alcohol. The biosensor was designed in a differential format consisting of a common Ag/AgCl reference electrode, an active working electrode containing the ADH, and the inactive working electrode containing only bovine serum albumin instead of the ADH. The differential signal between the active working electrode and the inactive working electrode minimized the interference from a large number of oxidizable species present in a person's breath. Prior to the amperometric measurement the biosensor was hydrated simply by dipping it into a phosphate buffer solution at pH 7.4. The NADH produced from the enzymatic reaction was oxidized at the working electrode biased at a potential of 470 mV vs. an on-board Ag/AgCl reference electrode. The biosensor can measure a person's breath alcohol over the concentration range 20±800 ppm routinely required in a test of drunken driving. # 1999 Elsevier Science B.V. All rights reserved.

Amperometric ethanol biosensor based on poly(vinyl alcohol)-multiwalled carbon nanotube-alcohol dehydrogenase biocomposite.

A novel amperometric ethanol biosensor was constructed using alcohol dehydrogenase (ADH) physically immobilized within poly(vinyl alcohol)-multiwalled carbon nanotube (PVA-MWCNT) composite obtained by a freezing-thawing process. It comprises a MWCNT conduit, a PVA binder, and an ADH function. The measurement of ethanol is based on the signal produced by beta-nicotinamide adenine dinucleotide (N...

Simple and Rapid Determination of Ethanol Content in Beer Using an Amperometric Biosensor

An alcohol dehydrogenase-based biosensor was prepared and tested for its use to determine ethanol in beer. The biosensor is based on a screen-printed carbon electrode (SPCE) modified by rhodium dioxide and immobilized with a biocatalytic layer containing the enzyme. Function of the enzyme biosensor was tested in model ethanol samples, in which it showed a linear range of 15–120 g∙L with a detec...

Amperometric biosensing of ethanol based on integration of alcohol dehydrogenase with a Pt/PPy–PVS/MB electrode

A novel amperometric ethanol biosensor with alcohol dehydrogenase (ADH) and nicotinamide adenine dinucleotide (NAD) on polypyrrole–polyvinylsulfonate (PPy-PVS) film was accomplished. Meldola’s blue (MB) was used as a mediator; it can facilitate the electron transfer that involves a chemical interaction between the electrode surface and NADH. Determination of ethanol was carried out by the oxida...

Computational Modeling of Mediator Oxidation by Oxygen in an Amperometric Glucose Biosensor

In this paper, an amperometric glucose biosensor is modeled numerically. The model is based on non-stationary reaction-diffusion type equations. The model consists of four layers. An enzyme layer lies directly on a working electrode surface. The enzyme layer is attached to an electrode by a polyvinyl alcohol (PVA) coated terylene membrane. This membrane is modeled as a PVA layer and a terylene ...

Detection of Waterborne and Airborne Formaldehyde: From Amperometric Chemosensing to a Visual Biosensor Based on Alcohol Oxidase

A laboratory prototype of a microcomputer-based analyzer was developed for quantitative determination of formaldehyde in liquid samples, based on catalytic chemosensing elements. It was shown that selectivity for the target analyte could be increased by modulating the working electrode potential. Analytical parameters of three variants of the amperometric analyzer that differed in the chemical ...
